体外研究

Mirtazapine对克隆的人α2A-肾上腺素(AR)受体表现出显著的亲和力，在该受体上其阻断去甲肾上腺素(NA)诱导的鸟苷-5'-O-(3-[35S]硫代)-三磷酸盐 ([35S]-GTPgammaS)结合的激活。Mirtazapine对克隆的人血清素(5-HT)2C受体表现出高亲和力，在该受体上其废除5-HT诱导的磷酸肌醇产生。Mirtazapine显著提高NA和DA在FCX中的渗透水平，而5-HT不被影响。[1] Mirtazapine通过阻断α2肾上腺素自身-和异型-受体，增强上升5-HT通路的电刺激疗效。Mirtazapine阻断微粒子电渗法施加的去甲肾上腺素(NE)对CA3背侧海马椎体神经元放电活性的抑制作用，表明其对突触后α2受体具有拮抗作用。[2]

体内研究

Mirtazapine (10-250 mg/kg i.v.)剂量依赖性增强天然大鼠中5-HT神经元的放电活性，但对6-羟多巴胺预处理的大鼠没有作用。[2]在雄性Sprague-Dawley大鼠中，Mirtazapine (5 mg/kg/day, s.c., 使用皮下埋置渗透性微泵)增加蓝斑核去甲肾上腺素(NA)神经元的自发放电活性。对于背侧海马CA3锥体神经元放电活性的抑制，α2肾上腺素受体激动剂可乐定对上升的5-HT通路电刺激产生作用，Mirtazapine在低剂量(10 mg/kg, i.v.)下拮抗强化作用，在高剂量(100 mg/kg, i.v.)下拮抗减弱作用。[3] Mirtazapine (5 mg/kg s.c.)仅轻微影响纹状体中DOPAC和高香草酸水平，几乎不影响自由活动大鼠中5-HT的释放，但是显著增加5-羟基吲哚乙酸。[4]

Mirtazapine is a noradrenergic and specific serotonergic antidepressant (NaSSA). Mirtazapine agonizes selective adrenergic and serotonergic receptors so that both NE release and 5-HT_1A mediated serotonergic signaling are increased.
