产品描述 Azelastine HCl是有效的，第二代选择性组胺拮抗剂。 靶点（IC50 & Targe） H1 receptor 体外研究 在大鼠肥大RBL-2H3细胞系中，Azelastine抑制Ag和ionomycin诱导的TNF-alpha释放，IC50值分别为25.7 mM和1.66 mM。在Ag刺激的细胞中，Azelastine也抑制TNF-α的mRNA表达和TNF-α蛋白的合成和释放，并可能涉及到这些效果，Ca 2+内流。Azelastine抑制TNF-α的释放，以比在离子霉素刺激的细胞中的mRNA表达/蛋白质合成和Ca 2+内流更大，这表明Azelastine通过用信号而不是钙离子抑制TNF-α的转录和产生的。Ionomycin刺激后加入Azelastine1小时后立即阻断TNF-α的释放，而不影响钙离子的内流。Azelastine抑制ionomycin诱导的，而不是Ag诱导的蛋白激酶C至细胞膜。[1] 在人牙龈成纤维细胞中，Azelastine盐酸（Azeptin）剂量依赖性地抑制了DNA和蛋白质的合成，也抑制人外周血淋巴细胞（PBL）的胚细胞。在小鼠腹腔巨噬细胞中，Azelastine盐酸（Azeptin）抑制诱导型一氧化氮合酶mRNA表达水平和NO的生成。[2] 在正常的人类肥大细胞中，Azelastine抑制IL-6，TNF-α和IL-8的分泌和NF-κB的活化，以及细胞内钙离子水平。[3] 参考文献 [1] Hide I, et al.
