Silodosin是一种α1-肾上腺素受体拮抗剂，具有高泌尿选择性。

体外研究

Silodosin比tamsulosin盐酸, naftopidil或prazosin盐酸有α（1A）-AR亚型更高的选择性 （亲和力tamsulosin盐酸最高，接着silodosin, prazosin盐酸或naftopidil）。[1]在人和大鼠离体前列腺中，Silodosin和tadalafil具有对大鼠神经介导的收缩效应的协同抑制。[2]

体内研究

在兔子的下泌尿道组织中，Silodosin强烈拮抗去甲肾上腺素诱导的收缩（包括前列腺，尿道和膀胱三角，PA（2）或pK b值分别为9.60，8.71和9.35）。[1] 在大鼠中，Silodosin在口服给药12小时，18小时和24小时后。显著抑制phenylephrine诱导的尿道内压力的增加（相对于对照处理组）。[3] Silodosin（0.1-0.3毫克/千克）或Prazosin（0.03-0.1毫克/公斤）减小障碍诱导腔内输尿管压力分别增加21-37％或18-40％。Silodosin抑制分离的大鼠和人输尿管收缩，有极好的功能选择性以减轻负荷大鼠阻塞输尿管的压力。[4] 在前列腺增生症的年轻和年老的狗中，Silodosin（0.3-300毫克/千克）剂量依赖性地抑制腹下神经刺激诱发的尿道压力增加（无显著降压效果）。[5]
