Zolmitriptan 是一种选择性5-羟色胺受体激动剂。

体外研究

在灵长类动物基底动脉和人的心外膜冠状动脉环中，Zolmitriptan产生浓度依赖性收缩。在转染的CHO-K1细胞膜中，Zolmitriptan显示人类重组5-HT1D（前5-HT1D alpha）和5-HT1B（前5-HT1D beta）受体的高亲和力。[1] 在表达重组人5-HT（1B）受体的C6胶质瘤细胞中，Zolmitriptan以浓度依赖的方式增加I(K)（最大增加16.3％），pD(2) 值为7.03。在表达克隆的人5-HT（1B）受体的C6细胞中，Zolmitriptan诱导的I(K)为钙离子螯合剂EGTA（5 mM）所抑制。[2]

体内研究

在麻醉豚鼠同侧硬脑膜中，Zolmitriptan（3-30 毫克/千克，静脉注射）在三叉神经节的单方面电刺激前十分钟给药剂量依赖性抑制[125I]血浆蛋白外渗。[1] Zolmitriptan（10-1000 毫克/千克，静脉注射）选择性降低动静脉-吻合（AVA）电导产生最大降低达到92.5%。Zolmitriptan也产生脑外电导的适度减少（最大减少23.9％，30 毫克/公斤，静脉注射），但不影响大脑电导的影响。在麻醉猫中，Zolmitriptan（1-30 毫克/千克，静脉注射）在耳微血管电导中产生剂量依赖性降低，它降低颈动脉微血管电导。[3] 在小鼠中，Zolmitriptan发挥行为上特异性抗攻击性的影响。在小鼠中，Zolmitriptan也降低酒精加强攻击性。[4]

Zolmitriptan is a selective serotonin receptor agonist of the 5HT1B and 5HT1D subtypes, both centrally and peripherally. It has been used clinically for the acute treatment of migraine attacks with or without aura and cluster headaches.
